The Leica Stellaris 5 LIAchroic Cryo is a cutting-edge Cryo-Correlative light and electron microscopy (Cryo-CLEM) solution integrated into the Cryo-electron tomography (Cryo-ET) workflow at the University Research Facility in Life Sciences (ULS). While a Cryo-Electron Microscopy (Cryo-EM) can visualize molecules at near-atomic resolution, it has a very narrow field of view and cannot locate a specific, rare biological event or specific protein identities on its own. Cryo-CLEM solves this by combining the functional fluorescence imaging and high-resolution structural analysis, allowing researchers to pinpoint specific cellular events under cryogenic conditions before proceeding to high-resolution imaging on our Cryo-TEM systems. The Stellaris 5 Cryo-CLEM system allows confocal imaging at cryogenic temperatures to identify precise coordinates of targets in vitrified biological specimen. By integrating this workflow, the users are provided with significantly increasing throughput and success rates for targeted Focused Ion Beam (FIB) lamella milling or structural data collection, streamlining the in situ cellular Cryo-ET workflow.
